Isoprostanoids are a group of non-enzymatic oxygenated metabolites of polyunsaturated fatty acids. It belongs to oxylipins group, which are important lipid mediators in biological processes, such as tissue repair, blood clotting, blood vessel permeability, inflammation and immunity regulation. Recently, isoprostanoids from eicosapentaenoic, docosahexaenoic, adrenic and α-linolenic namely F3-isoprostanes, F4-neuroprostanes, F2-dihomo-isoprostanes and F1-phytoprostanes, respectively have attracted attention because of their putative contribution to health. Since isoprostanoids are derived from different substrate of PUFAs and can have similar or opposing biological consequences, a total isoprostanoids profile is essential to understand the overall effect in the testing model. However, the concentration of most isoprostanoids range from picogram to nanogram, therefore a sensitive method to quantify 20 isoprostanoids simultaneously was formulated and measured by liquid chromatography-tandem mass spectrometry (LC–MS/MS). The lipid portion from various biological samples was extracted prior to LC–MS/MS evaluation. For all the isoprostanoids LOD and LOQ, and the method was validated on plasma samples for matrix effect, yield of extraction and reproducibility were determined. The methodology was further tested for the isoprostanoids profiles in brain and liver of LDLR−/− mice with and without docosahexaenoic acid (DHA) supplementation. Our analysis showed similar levels of total F2-isoprostanes and F4-neuroprostanes in the liver and brain of non-supplemented LDLR−/− mice. The distribution of different F2-isoprostane isomers varied between tissues but not for F4-neuroprostanes which were predominated by the 4(RS)-4-F4t-neuroprostane isomer. DHA supplementation to LDLR−/− mice concomitantly increased total F4-neuroprostanes levels compared to F2-isoprostanes but this effect was more pronounced in the liver than brain.  相似文献

It is shown that theC-metric (with parameters chosen to lie in suitable intervals) admits a conformal completion such that the space of generators of null infinity, , is a 2-sphere. This structure of is both necessary and sufficient for the analysis of gravitational radiation in exact general relativity. Bondi news (as well as the electromagnetic radiation field, in the charget case) is examined and found to be non-zero. Thus the issue of existence of exact solutions to the Einstein (and Einstein-Maxwell) equations admitting radiation (in the sense of Bondi, Sachs, and Penrose) is resolved. In addition, the analysis clarifies the sense in which the vacuumC-metric represents the gravitational field of two accelerating black-holes.  相似文献

Tyrosine sulfation is a post‐translational modification of many secreted and membrane‐bound proteins. It governs protein‐protein interactions that are involved in leukocyte adhesion, hemostasis, and chemokine signaling. However, the intrinsic feature of sulfated protein remains elusive and remains to be delineated. This investigation presents SulfoSite, which is a computational method based on a support vector machine (SVM) for predicting protein sulfotyrosine sites. The approach was developed to consider structural information such as concerning the secondary structure and solvent accessibility of amino acids that surround the sulfotyrosine sites. One hundred sixty‐two experimentally verified tyrosine sulfation sites were identified using UniProtKB/SwissProt release 53.0. The results of a five‐fold cross‐validation evaluation suggest that the accessibility of the solvent around the sulfotyrosine sites contributes substantially to predictive accuracy. The SVM classifier can achieve an accuracy of 94.2% in five‐fold cross validation when sequence positional weighted matrix (PWM) is coupled with values of the accessible surface area (ASA). The proposed method significantly outperforms previous methods for accurately predicting the location of tyrosine sulfation sites. © 2009 Wiley Periodicals, Inc. J Comput Chem, 2009  相似文献

A quantization procedure is given for the scalar field on stationary, axisymmetric background spacetimes with orthogonal 2-surfaces. The procedure is based on observers orthogonal to surfaces of constant Killing time, and thus agrees with the usual procedure for static spacetimes. For stationary but nonstatic spacetimes the procedure differs from the usual one but nonetheless leads to a natural quantization scheme. Applying the procedure to flat space in rotating coordinates gives the standard, inertial Minkowski vacuum. For the Kerr spacetime, the procedure yields a particle definition which is well-defined everywhere outside the horizon. The above observers are just nonrotating ZAMO's, and the vacuum state smoothly interpolates between the “in” and “out” Boulware vacua.  相似文献

Anderson and DeWitt considered the quantization of a massless scalar field in a spacetime whose spacelike hypersurfaces change topology and concluded that the topology change gives rise to infinite particle and energy production. We show here that their calculations are insufficient and that their propagation rule is unphysical. However, our results using a more general propagation rule support their conclusion.  相似文献
